Salmonella bacteria helped reduce tumor growth and prevent metastases

Salmonella is a pathogenic bacteria that can cause salmonellosis, a disease accompanied by

diarrhea in humans. Despite this, they can still penetrate the cell, parasitize it and multiply in special vacuoles.

Penetration into a cell is a complex process,which requires the generation of several types of protein. The authors of the new work decided to use this property of salmonella to deliver drugs directly to the tumor.

To do this, they modified a regulatory proteinflhDC, which is responsible for the synthesis of components for cell penetration. Biologists modified the bacteria in such a way that their flhDC levels were increased. 

In addition, so that the pathogenic bacterium does notdamaged tissue health, the authors of the work used a special SPI2 promoter, a site in bacterial DNA that activates certain genes. It only works inside the cell. Next, the authors placed next to it the genes responsible for the self-destruction of the bacterial cell.

At the final stage of the work, the researchers forcedbacteria synthesize a certain substance by adding the necessary genes. The safety of the system was tested on mice—the drug did not accumulate in healthy organs.

The authors also tested the new system on active ingredients: as a result, the NIPP1-CD and CT Casp-3 proteins slowed down the growth of tumors and prevented the onset of metastases in mice.
